What is Kombiglyze?
Kombiglyze XR is a prescription medication that helps adults with type-2 diabetes control blood-sugar levels. It contains a combination of saxagliptin (Onglyza) and metformin. Saxagliptin is a new type of diabetes drug made by AstraZeneca that has been on the market since 2009.
What is the problem?
People with diabetes have an increased risk of diabetic heart disease, which can lead to heart attacks and heart failure. This is why experts are so concerned about studies linking diabetes medications like saxagliptin with even higher rates of heart failure and death.
Kombiglyze Linked to Heart Failure and Death
Saxagliptin was associated with a 27% increased risk of hospitalization for heart failure in the SAVOR clinical trial, which was published in the New England Journal of Medicine in February 2014.
After the FDA reviewed results of the study, the agency concluded that the label should be updated to include warnings about heart failure. The FDA also warned about a “significant or near-significant” increased risk of death.
Conclusions of the study were based on data from 16,500 patients who were randomly assigned a placebo or saxagliptin. After about two years, 2.8% of placebo patients were hospitalized with heart failure, compared to 3.5% of patients on saxagliptin.
What is Heart Failure?
Heart failure occurs when the heart muscle is no longer strong enough to pump oxygen-rich blood into the body. Symptoms often begin slowly and gradually worsen as the heart muscle thickens, loses flexibility, and weakens.
Heart failure symptoms may include:
- Shortness of breath
- Persistent cough or wheezing
- Edema (fluid accumulation and swelling in tissues)
- Fatigue and weakness
- Nausea and loss of appetite
- Confusion
- Rapid heart rhythm
