Does Janumet Cause Pancreatic Cancer?

Researchers do not know if Janumet causes pancreatic cancer. Janumet pancreatic cancer is difficult to study because pancreatic cancer is very rare and Janumet has only been sold since 2007. The FDA has asked Amylin Pharmaceuticals, the drug-maker behind sitagliptin, to conduct a long-term study of sitagliptin and pancreatic cancer. Studies are currently underway, it will likely take many years before they are complete.

Adequate studies of Janumet pancreatic cancer are difficult because people who take Janumet often have multiple risk factors for pancreatic cancer — diabetes, pancreatitis, age, gender, etc. It can be hard to say whether a case of pancreatic cancer was caused by Janumet or another risk factor. However, there is evidence linking Janumet to pancreatic cancer:

  • In 2011, Gastroenterology published a study from researchers at UCLA who identified 16 reports of pancreatic cancer in sitagliptin patients between 2004 – 2009.
  • Janumet was linked to 88 cases of acute pancreatitis in an FDA warning letter in 2009. Pancreatitis is a risk factor for developing pancreatic cancer.

Pancreatic Cancer Causes

All cancers are caused by cells that mutate and begin multiplying out of control. However, it is unknown what causes these mutations. Pancreatic cancer can be caused by endocrine (hormone-making) or exocrine (enzyme-making) cells in the pancreas. Endocrine pancreatic cancer is rarer than exocrine pancreatic cancer, but the prognosis is better.

Risk factors that may cause pancreatic cancer include:

  • Janumet: This drug and other GLP-1 therapy drugs specifically target pancreatic cells.
  • Pancreatitis: Inflammation in the pancreas that can be chronic or acute
  • Age: Nearly all cases of pancreatic cancer occur in people over 50 years old, and the risk increases with age.
  • Smoking cigarettes
  • Diabetes
  • Obesity
  • Liver problems: Chronic liver diseases, such as cirrhosis of the liver
  • Diet: Some research suggests that diets high in fat, fried foods, processed and/or smoked meats increase the risk of causing pancreatic cancer.
  • Alcoholism
  • Occupational exposure: Dyes, chemicals used in metal refining, and pesticides.
