FDA Evaluating Rhabdomyolysis Risk

The FDA is evaluating a potential signals of a serious risk and new safety information linking DPP4 diabetes drugs and rhabdomyolysis. This potentially life-threatening kidney side effect was identified by the FDA Adverse Event Reporting System (FAERS) between April and June 2017.

Oseni and Pancreatic Cancer

Oseni is a combination of pioglitazone (Actos) and alogliptin (Nesina). These drugs treat type-2 diabetes in different ways. Oseni stimulates the pancreas to make more insulin, which helps lower blood-sugar levels.

Because Oseni relies so heavily on the pancreas and people with diabetes already have a higher risk of pancreas problems, many experts are concerned about side effects, such as:

  • Chronic pancreatitis
  • Hemorrhagic and necrotizing pancreatitis
  • Pancreatic cancer

FDA Investigates Studies of Pancreatic Side Effects

In March 2013, the FDA warned that two other drugs in the same class as Oseni — sitagliptin and exenatide — had been linked to a doubled increased risk of pancreatitis in a recent study. Pancreatic cancer was also more commonly reported in patients on sitagliptin or exenatide, according to a study published in Gastroenterology in 2010.

Pancreatitis and Pancreatic Cancer

Long-term inflammation in the pancreas (chronic pancreatitis) is a risk-factor for pancreatic cancer. Over time, inflammation causes more cells to divide and grow, which increases the odds of a mutation that causes cells to grow uncontrollably.

Why Is Pancreatic Cancer So Dangerous?

Most types of pancreatic cancer occur in exocrine cells, which are responsible for making digestive enzymes. These tumors usually grow aggressively, spread to several vital organs (metastasize) near the pancreas, and produce few symptoms in the early stages. This “perfect storm” is why pancreatic cancer is one of the deadliest cancers, despite being so rare.
